These variants, though uncommon, have a powerful effect on genes expressed in brain nerve cells. This disruption in the development and communication between these cells can contribute to ADHD. The study's senior author, Professor Anders Børglum, explains that for the first time, they can pinpoint specific genes where rare variants significantly elevate the risk of ADHD.

These findings suggest that these variants likely have a highly damaging effect on the genes, revealing which genes and fundamental biological mechanisms may be affected.

But here's where it gets controversial... the study also showed that these effects can be traced back to fetal life and continue into adulthood. The researchers found that the rare variants particularly affect the function of dopaminergic and GABAergic neurons, which help regulate attention, impulse control, and motivation. This supports the idea that disturbances in brain development and function are central to ADHD.

Furthermore, the research identified a larger protein network that interacts with the proteins encoded by the three identified ADHD genes. And this is the part most people miss... the impact of these rare genetic variants extends beyond just who develops ADHD. The study revealed that individuals with these variants tend to have lower educational attainment and socioeconomic status. For adults with ADHD, an average reduction in IQ score of around 2.25 points was observed for each rare high-risk variant they carry.

This suggests that cognitive challenges are more pronounced in individuals with ADHD due to these rare genetic variants, potentially leading to educational and occupational consequences.

'This is only the beginning,' says Jinjie Duan, a postdoctoral researcher in the Aarhus group. The findings broaden our understanding of the biological underpinnings of ADHD and may pave the way for future treatments. Researchers are already pointing to 17 additional genes with rare variants that are likely causal.
